design and fabrication of anatomical bioreactor systems containing alginate scaffolds for cartilage tissue engineering

چکیده

the aim of the present study was to develop a tissue-engineering approach through alginate gel molding to mimic cartilage tissue in a three-dimensional culture system. the perfusion biomimetic bioreactor was designed to mimic natural joint. the shear stresses exerting on the bioreactor chamber were calculated by computational fluid dynamic (cfd). several alginate/bovine chondrocyte constructs were prepared, and were cultured in the bioreactor. histochemical and immunohistochemical staining methods for the presence of glycosaminoglycan(gag), overall matrix production and type ii collagen protein were performed, respectively. the dynamic mechanical device applied a linear mechanical displacement of 2 mm to 10 mm. the cfd modeling indicated peak velocity and maximum wall shear stress were 1.706×10-3 m/s and 0.02407 dyne/cm2, respectively. histochemical and immunohistochemical analysis revealed evidence of cartilage-like tissue with lacunas similar to those of natural cartilage and the production of sulfated gag of matrix by the chondrons, metachromatic territorial matrix-surrounded cells and accumulation of type ii collagen around the cells. the present study indicated that when chondrocytes were seeded in alginate hydrogel and cultured in biomimetic cell culture system, cells survived well and secreted newly synthesized matrix led to improvement of chondrogenesis.

Chitosan scaffolds containing hyaluronic acid for cartilage tissue engineering.

Scaffolds derived from natural polysaccharides are very promising in tissue engineering applications and regenerative medicine, as they resemble glycosaminoglycans in the extracellular matrix (ECM). In this study, we have prepared freeze-dried composite scaffolds of chitosan (CHT) and hyaluronic acid (HA) in different weight ratios containing either no HA (control) or 1%, 5%, or 10% of HA. We h...

design and fabrication of biodegradable porous chitosan/gelatin/tricalcium phosphate hybrid scaffolds for tissue engineering

in this study, based on a biomimetic approach, novel 3d biodegradable porous hybrid scaffolds consisting of chitosan, gelatin, and tricalcium phosphate were developed for bone and cartilage tissue engineering. macroporous chitosan/ gelatin/β-tcp scaffolds were prepared through the process of freeze-gelation/solid-liquid phase separation. the results showed that the prepared scaffolds are highly...

Fabrication of Polymer Scaffolds for Tissue Engineering

A new approach to the fabrication of individual implants and scaffolds for tissue engineering—surface selective laser sintering (SSLS)—is proposed and realized. In contrast to the conventional selective laser sintering, the SSLS method makes it possible to sinter polymer microparticles and melt the near-surface layer rather than the microparticle as a whole. The effect of the laser radiation pa...

Fabrication of Porous Hydroxyapatite-Gelatin Composite Scaffolds for Bone Tissue Engineering

Background: engineering new bone tissue with cells and a synthetic extracellular matrix represents a new approach for the regeneration of mineralized tissues compared with the transplantation of bone (autografts or allografts). Methods: in this study, to mimic the mineral and organic component of natural bone, hydroxapatite (HA) and gelatin (GEL) composite scaffolds were prepared. The raw mater...

Fabrication of Porous Hydroxyapatite-Gelatin Scaffolds Crosslinked by Glutaraldehyde for Bone Tissue Engineering

In this study, to mimic the mineral and organic components of natural bone, hydroxyapatite[HA] and gelatin[GEL] composite scaffolds were prepared using the solvent-casting method combined with a freeze drying process. Glutaraldehyde[GA] was used as a cross linking agent and sodium bisulfite was used as an excess GA discharger. Using this technique, it is possible to produce scaffolds with mecha...
